Zusammenfassung
Auf der Grundlage eines etablierten physiologisch orientierten Modellierungsansatzes für toxikokinetische Erscheinungen flüchtiger Substanzen im Säugetierorganismus ist eine toxikokinetische Modell- und Datenbank su entwerfen, zu implementieren und unter besonderer Berücksichtigung der Toxikokinetik des Bensens su validieren. Eine Bestandsaufnahme der in der Literatur vorhandenen Informationen (Struktur, Prosesse, Daten der Modelle) führt sur Schaffung eines Objektwissen integrierenden Arbeitsmittels in der Hand der experimentell tätigen Toxikologen. Die physiologisch orientierte Struktur des Transportsystems ist durch ein die Metabolisierung beschreibendes Subsystem ergänzt. Dieses Hybridmodell kann sur Interpretation und Bewertung von Bildungsraten toxischer Metaboliten benutzt werden. Bei der Entwicklung werden neben dem Biomonitoring (exponierter Arbeiter) vor allem die Anforderungen tierexperimenteller Toxisitätsbewertung berücksichtigt. Der Modellierungsansats erlaubt Spezies-Extrapolationen und kann so entscheidend zur Verbesserung der Toxizitätsbewertung beitragen. Hier sind die entsprechenden Spezies-bezogenen Daten zu beschaffen und einer kritischen Bewertung zu unterziehen.
