Zusammenfassung
In diesem Artikel wird eine Methode zur Auflösungsverbesserung in der Fluoreszenzmikroskopie mit Hilfe der Rekonstruktion von Aufnahmen aus verschiedenen Blickrichtungen (Tomographie) vorgestellt. Der theoretisch erreichbare Auflösungsgewinn wird analytisch berechnet und an einem Fallbeispiel erläutert. Da-im Gegensatz zur Computertomographie — die Rotationsgeometrie der Tomographie nicht aus dem apparativen Aufbau selbst bestimmt werden kann, wird ein Alignmentalgorithmus vorgestellt, der zwei 3D Bildserien, die aus verschiedenen Blickwinkeln aufgenommen wurden, in ein Koordinatensystem einbettet. Mit Hilfe eines Vorverarbeitungsschritts, der Isotropfaltung, wird die Sensibilität des Verfahrens bzgl. der anisotropen Punktantwortfunktion des Fluoreszenzmikroskops reduziert.
