ANOVA Statistical Significance Analysis of Clinicopathological Characteristics Tumor Tissue

Objective: The aim of this study was to evaluate association of circulating and tumor tissue miR-141-5p expressions with the clinicopathological characteristics and overall survival (OS) in surgical patients with ICC. Methods: In this prospective study, 77 surgical patients with ICC were enrolled. Peripheral blood sample was collected the day before the operation, and ICC tissue specimens were obtained during the surgery. MiR-1415p expression was determined by quantitative polymerase chain reaction (qPCR). The median follow-up was 25.0 months, and last follow-up was June 30, 2017. Results: MiR-141-5p expressions in tumor tissue were positively associated with which in the peripheral blood mononuclear cell (PBMC) (R = 0.350,P = 0.002). The expression level of tissue miR-141-5p was negatively associated with N stage (P = 0.014). However, the expression level of PBMC miR-141-5p had no correlation with any clinicopathological characteristics of ICC patients. Kaplan-Meier curves indicated that high expression of tissue miR-141-5p (P < 0.001) was correlated with longer OS, while PBMC miR-141-5p didn't (P = 0.573). Univariate Cox regression analysis showed that tissue miR-1415p could predict better OS (P < 0.001), while ECOG high performance (P = 0.009), T high stage (P < 0.001), N high stage (P = 0.010), M high stage (P = 0.049) and TNM high stage (P = 0.003) could predict poor OS. Furthermore, multivariate Cox regression analysis revealed that only tissue miR-141-5p (P = 0.001) high expression was an independent factor for prolonged OS, and T high stage was a risk factor for OS. Conclusion: Tumor tissue miR-141-5p expressions were negatively associated with N stage of ICC, while PBMC miR-141-5p had no correlation with any clinicopathological characteristics of ICC patients. And tissue miR-141-5p could act as a novel and convincing prognostic biomarker in surgical patients with ICC.
