Zusammenfassung
Ein stetiges deterministisches mathematisches Modell der Erythropoese wurde entwickelt, das alle Zelltypen der erythropoetischen Zellinie enthält von Stammzellen bis zu Erythrozyten. Das Modell stellt sich als System von über 70 nicht linearen Differentialgleichungen dar und kann deshalb nur auf einer Rechenanlage simuliert werden. Experimentelle Daten zur Erythropoese der Maus wurden in eigenen Labors bestimmt oder der Literatur entnommen. Neben Daten des Gleichgewichtszustands wurden auch Meßwerte nach Blutung, Hypertransfusion, Injektion von Erythropoetin oder Hydroxyurea, Bestrahlung oder Kombinationsbehandlung herangezogen. Die Modellparameter ließen sich derart anpassen, daß das Modell alle genannten Situationen hinreichend gut nachvollziehen konnte. Die Regulation im Modell beruht auf Zell-zu-Zell Interaktionen und dem Hormon Erythropoetin (EPO). Trotz der gebotenen Vorsicht bei der Interpretation von Modellergebnissen und trotz weniger widersprüch¬licher Daten, scheint es außer EPO keinen weiteren humoralen Regulationsfaktor der Erythropoese zu geben.
Summary
A deterministic continuous mathematical model of erythropoiesis was developed which incorporates all cell types of the erythropoietic Line from stem cells to erythrocytes. The model is represented by a system of some 70 non linear differential equations and therefore can only be simulated on a Computer. Experimental data concerning the erythropoiesis of mice (CFU-S, BFU-E, CFU-E, iron incorporation) were determined in our laboratories or extracted out of the literature. The data include steady-state values as well as measurements after bleeding, hypertransfusion, injection of erythropoietin or hydroxyurea, irradiation, or combinations of these treatments. The parameters of the model were adjusted such that the model could reproduce all of these different situations reasonably well. Assuming short-range influences (cell-to-cell interactions) and the hormone erythropoietin (EPO) as regulators the model was used to test whether some other humoral factors besides EPO have to be assumed. Notwithstanding difficulties in the interpretation of modelling results and besides some contradictory experimental data this seems not to be the case.
