Abstract
In pathological diagnosis, a tissue sample is usually observed under a microscope after the tissue has been stained, but the stain conditions and the characteristics of observation devices vary depending on hospitals. That makes objective or quantitative diagnosis to be more difficult. This paper proposes the method for quantification of stain conditions from microscopic image obtained as transmittance spectra that is independent of observation devices. In the proposed method, Beer-Lambert law is used to obtain the amount of stain color pigments in an arbitrary tissue sample using the absorbance spectra of the stain color pigments. Using the transmittance spectral image, which has been estimated from the signal of multispectral digital camera, we can estimate the feature parameters characterizing the stain conditions. Through the experiment using the Hematoxilyn & Eosin stained tissue samples, the effectiveness of the proposed method is confirmed.
