Comparison of HPTLC and HPLC to determine the content of apiin in Apium graveolens L. root and the greenness assessment

Abstract

Apiin is the main active ingredient in Xinjiang Apium graveolens L. root (AR), which can be assayed by high-performance liquid chromatography (HPLC). However, apiin analysis by high-performance thin layer chromatography (HPTLC) is lacking in the available literature. Therefore, a fast and convenient HPTLC method was established in this experiment to detect apiin in Xinjiang AR qualitatively and quantitatively. Simultaneously, the HPLC method was used to determine its content, and the greenness of the two methods was compared and evaluated. Methodological investigation results show that the two methods have a good linear relationship in the range of 0.3–1.8 μg, and the precision, repeatability, stability, and recovery rate meet the analytical requirements. Comparing the two analytical methods from the perspective of green chemistry, HPTLC consumes fewer reagents, time, and waste liquid production than HPLC. The evaluation results of Analytical Eco-Scale (AES) show that HPTLC is an excellent green analysis, while HPLC is an acceptable green analysis. The Green Analysis Program Index (GAPI) and Green Assessment Profile (GAP) found that HPTLC is a green and environmentally friendly analysis method.
