Molecular Mechanism of Wenyang Zhenshuai Granule in Treatment of Chronic Heart Failure via Network pharmacology

Abstract

Objective: To predict the potential pharmacological mechanism of Wenyang Zhenshuai Granule (WYZSG) in the treatment of chronic heart failure via the network pharmacological method. Methods:The active components of WYZSG were screened through TCMSP and other databases. The related targets of chronic heart failure were screened through Genecards, OMIM, Disgenet databases, etc. Cytoscape3.7.2 was used to construct the network of "drugs-active components-targets". And not only was the String used for protein-protein interaction network, but also the BisoGenet database was used to supplement. The intersection network was extracted and the key targets of WYZSG in the treatment of chronic heart failure were obtained. Finally, GO and KEGG enrichment analysis of key targets were carried out by using Metascape. Results: The active components of WYZSG were Quercetin, Pachymic acid, Salsolinol, 1-methyl-6,7-dihydroxy-3,4-dihydroisoquinoline monohydrate, Ginsenoside, Gomisin R, Stigmasterol, etc. There are 163 key targets. The key biological processes and pathway may include AGE-RAGE signaling pathway, HIF-1 signaling pathway, Fluid shear stress and atherosclerosis, TNF signaling pathway, etc. Conclusions: This study preliminarily revealed the mechanism of WYZSG in treating chronic heart failure with multi components, multi targets and multi pathways, that may be effected through the regulation of complement system, cytokines and MAPK signal pathway. WYZSG may improve the long-term prognosis of patients with chronic heart failure by interfering with the biological processes of myocardial cell response to ROS (reactive oxygen species), ROS metabolic process and apoptosis. Through the discussion and analysis of network pharmacology, it provided potential target and basis for further study of WYZSG in the treatment of chronic heart failure, and provided basis for clinical development and utilization of WYZSG.
