Genetic Diversity of 60 Oriental Tobacco Germplasm Resources Revealed by SSR Markers

Abstract

In order to explore, identify, innovate and utilize of oriental tobacco germplasm resources, the genetic diversity of 60 oriental tobacco germplasms from 11 different countries was first analyzed by 35 SSR markers which evenly distributed on 24 linkage groups of tobacco. The results showed that 447 genetic loci existed in 60 oriental tobacco germplasms using 35 SSR primer pairs. The average observed heterozygosity (Ho) was 0.6231, and the average expected heterozygosity (He) was 0.708 2. The average Shannon's information index (I=1.415 9), the average Nei's diversity index (H=0.694 0) and the range of genetic distance (0.009∼0.924) were relatively high, which indicated that the genetic variation of 60 materials was quite large and the genetic diversity was much abundant in oriental tobacco. The result of cluster analysis revealed that 60 materials could be clustered into 3 groups at the level of GD=0.485, and which was highly correlated with their geographical origins.
